Toby Alderweireld's injury is not thought to be serious, according to manager Mauricio Pochettino, but Tottenham still wait on the results of a scan.
Alderweireld had to be stretchered off after colliding with team-mate Jan Vertonghen in the second half of Spurs' 1-1 draw at West Brom on Saturday.
Alderweireld appeared in serious pain and received lengthy treatment before attempting, but failing, to carry on in the game and Pochettino admitted it was a "heavy knock" to his right leg.
He has not travelled as part of Spurs' Champions League squad to face Bayer Leverkusen on Tuesday but Pochettino said: "We believe it is not a big issue.
"Now we have to wait for the result of the scan, which he took this afternoon. Everyone else is okay. We are only missing Harry Kane."
Eric Dier replaced Alderweireld at centre-back at the Hawthorns and he is set to continue partnering Vertonghen in Germany, with Pochettino suggesting Cameron Carter-Vickers is too inexperienced for such a significant game.
On Dier, Pochettino added: "The important thing is we trust in him. It's not important the position he will play - midfield, centre back or full back - we believe and trust in him.
"We trust him as a person and his skills. For me, Eric is a player who can develop his game in different positions."
Right-back Kyle Walker has not travelled and is being rested, with Kieran Trippier his natural replacement, and Pochettino said: "The next two games, here and Wembley, will decide the possibility to go to the next round
"It's important tomorrow that we win. We want to go to the next round, so it's key tomorrow to take the three points, then to play at Wembley in the same way."
