Louis van Gaal says neither Ander Herrera nor Phil Jones will be fit to face PSV - but hopes Wayne Rooney and Anthony Martial will be available.
United won 2-1 at Vicarage Road and briefly topped the Premier League despite making the trip south without six first-team players.
And Van Gaal lost two more ahead of Wednesday's Champions League game as Herrera and Jones had to be replaced either side of half-time.
Herrera set up United's opening goal for Memphis Depay only to injure his hamstring midway through the first half, and Jones departed with a twisted ankle after the break.
Van Gaal said: "We are in a lousy period because there are a lot of injuries.
"Jones and Herrera are not available for Wednesday. The first impression is they are not heavy injuries but we have to play Wednesday. Jones has a twisted ankle and Herrera maybe a hamstring injury."
Depay started as a central striker in the absence of Wayne Rooney and Anthony Martial, but both could be back against PSV as United chase the win which would secure qualification for the knockout stages.
"Rooney is ill and illness is a matter of days, not weeks," Van Gaal said. "I do expect that he shall play Wednesday. He is at home. Tomorrow he comes and then I can say more.
"Martial has a foot problem. It is not so heavy as the media has written but you never know, we have to wait and see."
