The 22-year-old former Alaves player's previous deal at San Mames, which he signed upon joining the club in 2009, had been due to expire next summer.
However, De Marcos has now extended his stay after agreeing a contract which sets his buy-out clause at 32millions euros.
The versatile player has enjoyed a more prominent role this season under new coach Marcelo Bielsa thanks to his ability to adapt to the Argentinian's myriad of different systems.
The deal is especially significant given the added importance of player retention at Athletic due to the ongoing Basque-only policy at the club.
