Ambler, a former Salford prop, has been included in the Rhinos' initial 19-man squad in place of England captain Jamie Peacock.
Peacock is out with sprained ankle ligaments, joining long-term casualties Kallum Watkins (knee), Scott Donald (shoulder), Greg Eastwood (wrist) and Carl Ablett (ankle) on the sidelines.
Leeds, however, are able to welcome back five players who missed the defeat to Hull KR last time out - Ryan Hall, Danny Buderus, Kevin Sinfield and Tom Bush have shaken off injuries and Ryan Bailey has recovered from illness
Cameron Phelps is expected to feature for Wigan after overcoming a thigh injury.
