Nathan Brown's men ran in 10 tries in a clinical display that leaves them guaranteed of a play-off place.
Stand-off Kevin Brown played a key role in three first-half tries as Huddersfield went on to break the half-century barrier against Wakefield for the second time this season.
The Giants beat Wakefield 52-0 in March and made it 110 points against the Wildcats with a 10-try blitz in front of their delirious home supporters.
Ruthless
David Hodgson kicked off the scoring after just eight minutes before Brown cheekily kicked through for Lee Gilmour to race over the line five minutes later.
Former Wakefield hooker Danny Brough, who kicked nine out of 10 conversion attempts against his old employers, then put Darrell Griffin away for a try before Huddersfield got a lucky break in the 26th minute and ruthlessly punished Wakefield with a quick counter-attack.
Scott Grix dropped a high kick near his own try-line as Huddersfield defended against some rare Wakefield pressure.
But referee Richard Silverwood allowed play to carry on and the Giants piled on the misery when Shaun Lunt broke down the right flank for a solo score.
The Wildcats went into half-time in disarray courtesy of two more Huddersfield tries in as many minutes. Brown conspired with Grix and then Jermaine McGillvary as the exciting youth product scored his seventh try in only five starts on 37 minutes.
Rout
Leroy Cudjoe then worked a brilliant one-two with McGillvary to leave Huddersfield with a 36-0 half-time lead.
Giants chairman Ken Davy told supporters at half-time that the Galpharm Stadium is to have a new pitch fitted next summer, meaning Huddersfield will play some games at Halifax next season.
Wakefield finally got off the mark early in the second half when Sean Gleeson dived over and Ben Jeffries converted.
But the Giants hit back and added further tries through Cudjoe, Grix, Brough and Graeme Horne before the final hooter. Brough completed the rout with three more conversions.
