The Cherry and Whites had made an impressive start at Kingsholm, scoring an early try through Freddie Burns before the two injuries occurred and both players had to be stretchered off.
"It's very unfortunate that they both have injuries to their lower legs and at this stage we don't know the extent of their problems," said Gloucester coach Andrew Stanley.
"We started off energetically in those first 20 minutes and created a few opportunities but those injuries and the break in play disrupted our flow."
Gloucester did eventually emphasise their superiority with a second-half try from Mike Tindall, while Burns kicked 10 points.
Rory Clegg's penalty was the Falcons' only contribution.
"Newcastle brought down a pretty strong side for this competition but after the interval we refocused and started again with one of the real plusses was keeping our line intact," Stanley added.
Newcastle director of rugby Dean Richards was unhappy with his side's performance.
"Our players are very despondent as they know they played with a lack of accuracy," he said. "We lacked a little bit of composure and turned over the ball far too often and played in the wrong areas of the field.
"It was great learning curve for some of our youngsters, especially front rowers, George McGuigan and Scott Wilson who are only 19 and 20 years of age."
