Jamahl Lolesi, Kevin Brown, Luke Robinson, Eorl Crabtree and Shane Elford all touched down for the Giants as they claimed their second successive victory after starting the season with back-to-back defeats.
Craig Hall scored the solitary try for injury-hit Hull, who have lost three and won only one of their opening four Super League matches.
Huddersfield had not beaten Hull in eight meetings, including last season's play-off at the KC Stadium, but they gained some revenge with a well-deserved success.
Early strike

Giants gradually cranked up the gas and grabbed the first four-pointer on 23
minutes, Lolesi touching down after a dazzling run from replacement hooker George Gatis.
Chris Foreman converted but Hull restored parity five minutes later, Craig Hall
finishing off a slick move involving Lee Radford, Danny Washbrook and Graeme Horne.
Tickle's conversion made it 8-6 to the visitors as the ex-Wigan goal-kicker
became the latest entrant to the 1000 Super League points club.
A brilliant ankle-tap from Matt Sing then denied a certain try for Rod Jensen
but the Giants' winger played a decisive hand moments before the break, setting up Brown, Foreman's goal making it 12-8 at the break.
Giants turned the screw on the restart and after Ryan Hudson was held just short, Robinson powered over from dummy-half for his fifth try in four matches this term, Foreman's conversion handing the hosts a 10-point buffer.
The hosts made the game safe when Crabtree crossed the whitewash shortly after a barnstorming run from Brown, who cashed in on a loose pass from Horne.
Elford put gloss on the win three minutes from time, diving over after
gathering Foreman's neat kick - the full-back converting.
